The Investigation of the Apoptose Structural Effects and Mechanism in Leukemic Cells of Sirt1 Inhibitor Sirtinol  [PDF]

Abstract:
Human SIRT1 is an enzyme that deacetylates the p53 tumor suppressor protein. It has been suggested to modulate p53-dependent functions including DNA damage-induced cell death. Sirtuins are nicotinamide adenine dinucleotide dependent class III histone deacetylase proteins that play a crucial role in several cellular processes, including DNA repair, apoptosis, and lifespan. In this study, we investigated the relationship between sirtinols and apoptosis mechanism at leukemic cells. For this, we applied sirtinol to K-562 (chronic myeloid leukemia) and Jurkat (acute T-cell leukemia) cell lines at different dilutions in cell culture conditions. And Cdna isolated patient RNA samples, after that Caspase3, Bax, Bcl2 expression performed with used qRT-PCR technique and cells were stained by Annexin V method. According to the results of research, we identified that Sirtinol dilutions were increased in both K-562 and Jurkat, while the number of living cells was decreasing, the number of dead cells increased at 50 μM dilution. Sirtinol, an HDAC inhibitor in the direction of these results, has been observed to be a drug that can be effective in the treatment of CML as well as T-ALL.

Abstract:
CD95 is a cell surface receptor involved in apoptotic signal transmission. Deregulation of this pathway results in down regulation of apoptosis and subsequent persistence of a malignant clone. A single nucleotide polymorphism (SNP) resulting in guanine-to-adenine (G > A) transition in the CD95 promoter region (position ﹣1377) is thought to reduce stimulatory protein 1 (SP1) transcription factor binding and decrease CD95 expression. The purpose of this study was to examine a genetic polymorphism in the core promoter of CD95 is associated with the frequency. G-1377A in the CD95 promoter region was genotyped by polymerase chain reaction and restriction endonuclease analysis and sequenced of the CD95 gene (Beckman Coulter, USA). Among 146 patients, CD95 G-1377A (rs2234767) SNP carriers frequencies has been identified as 25% GA (n:37) and AA 4% (n:6), respectively. This polymorphism of the distribution of the CD95 gene in acute leukemic children will be a guide for future studies.
Notes on Weak Units of Group-Like 1- and 2-Stacks

Abstract: The weak units of strict monoidal 1- and 2-categories are already defined. In this paper, we define them for group-like 1- and 2-stacks. We show that they form a contractible Picard 1- and 2-stack, respectively. We give their cohomological description which provides for these stacks a representation by complexes of sheaves of groups. Later, we extend the discussion to the monoidal case.
Extensions of Picard 2-Stacks and the cohomology groups Ext^i of length 3 complexes

Abstract: The aim of this paper is to define and study the 3-category of extensions of Picard 2-stacks over a site S and to furnish a geometrical description of the cohomology groups Ext^i of length 3 complexes of abelian sheaves. More precisely, our main Theorem furnishes (1) a parametrization of the equivalence classes of objects, 1-arrows, 2-arrows, and 3-arrows of the 3-category of extensions of Picard 2-stacks by the cohomology groups Ext^i, and (2) a geometrical description of the cohomology groups Ext^i of length 3 complexes of abelian sheaves via extensions of Picard 2-stacks. To this end, we use the triequivalence between the 3-category of Picard 2-stacks and the tricategory T^[-2,0](S) of length 3 complexes of abelian sheaves over S introduced by the second author in arXiv:0906.2393, and we define the notion of extension in this tricategory T^[-2,0](S), getting a pure algebraic analogue of the 3-category of extensions of Picard 2-stacks. The calculus of fractions that we use to define extensions in the tricategory T^[-2,0](S) plays a central role in the proof of our Main Theorem.
Resolution of extensions of Picard 2-stacks

Abstract: Let S be a site. First we define the 3-category of torsors under a Picard S-2-stack and we furnish (1) a parametrization of the equivalence classes of objects, 1-arrows, 2-arrows and 3-arrows of the 3-category of torsors under a Picard S-2-stack by the cohomology groups of the derived functor of the functor of global sections, and (2) a geometrical description of the cohomology groups of the derived functor of the functor of global sections applied to length 3 complexes of abelian sheaves via torsors under a Picard S-2-stack. Then we describe extensions of Picard S-2-stacks in term of torsors under a Picard S-2-stack which are endowed with a group law on the fibers. As a consequence of such a description, we get an explicit right resolution of the 3-category of extensions of Picard S-2-stacks in terms of 3-categories of torsors under a Picard S-2-stack. Using the dictionary between the derived category of abelian sheaves on S and the category of Picard S-2-stacks, we rewrite this categorical right resolution in homological terms.
